چکیده

We report the formation of lipid membranes supported by a soft polymeric cushion of polydopamine. First, 20 nm thick polydopamine films were formed on mica substrates. Atomic force microscopy imaging indicated that these films were also soft with a surface roughness of 2 nm under hydrated conditions. A zwitterionic phospholipid bilayer was then deposited on the polydopamine cushion by fusion of dimyristoylphosphatidylcholine (DMPC) and dioleoylphosphatidylcholine (DOPC) vesicles. Polydopamine films preserved the lateral mobility of the phospholipids as shown by fluorescence microscopy recovery after photobleaching (FRAP) experiments. Diffusion coefficients of ~5.9 and 7.2 μm s were respectively determined for DMPC and DOPC at room temperature, values which are characteristic of lipids in a free standing

Substrate Effects on the Formation Process, Structure and Physicochemical Properties of Supported Lipid Bilayers

Supported lipid bilayers are artificial lipid bilayer membranes existing at the interface between solid substrates and aqueous solution. Surface structures and properties of the solid substrates affect the formation process, fluidity, two-dimensional structure and chemical activity of supported lipid bilayers, through the 1–2 nm thick water layer between the substrate and bilayer membrane. Even...

Solid supported lipid bilayers: From biophysical studies to sensor design

The lipid bilayer is one of the most eloquent and important self-assembled structures in nature. It not only provides a protective container for cells and sub-cellular compartments, but also hosts much of the machinery for cellular communication and transport across the cell membrane. Solid supported lipid bilayers provide an excellent model system for studying the surface chemistry of the cell...

Formation and Characterization of Supported Lipid Bilayers Composed of Hydrogenated and Deuterated Escherichia coli Lipids

Supported lipid bilayers are widely used for sensing and deciphering biomolecular interactions with model cell membranes. In this paper, we present a method to form supported lipid bilayers from total lipid extracts of Escherichia coli by vesicle fusion. We show the validity of this method for different types of extracts including those from deuterated biomass using a combination of complementa...

Cell adhesion to protein-micropatterned-supported lipid bilayer membranes.

A new method for constructing controlled interfaces between cells and synthetic supported lipid bilayer membranes is reported. Microcontact printing is used to define squares and grid lines of fibronectin onto glass, which subsequently direct the self-assembly of fluid lipid bilayers onto the complementary, uncoated regions of the surface. Features of fibronectin as small as 5 microm effectivel...

Preparation of DOPC and DPPC Supported Planar Lipid Bilayers for Atomic Force Microscopy and Atomic Force Spectroscopy

Cell membranes are typically very complex, consisting of a multitude of different lipids and proteins. Supported lipid bilayers are widely used as model systems to study biological membranes. Atomic force microscopy and force spectroscopy techniques are nanoscale methods that are successfully used to study supported lipid bilayers. These methods, especially force spectroscopy, require the relia...
